Thermo Scientific™ Phusion™ Plus DNA Polymerase is a hot-start high quality DNA polymerase that combines protein fusion technology with universal primer annealing. Phusion Plus DNA Polymerase is a Pyrococcus-like proofreading DNA polymerase fused to a process-enhancing domain that enables PCR arrays to be generated with high accuracy, sensitivity and inhibitor tolerance.

Thermo Scientific™ Phusion™ Plus PCR Master Mix is a convenient, ready-to-use 2X mix of Phusion Plus DNA Polymerase, buffer, magnesium chloride and dNTPs. The master mix format simplifies reaction setup with minimal pipetting steps and retains all the features of the standalone format.

Thermo Scientific™ Phusion™ Plus Green PCR Master Mix is a convenient, ready-to-use 2X mix of Phusion Plus DNA Polymerase, buffer, magnesium chloride and dNTPs. The master mix format simplifies reaction setup with minimal pipetting steps and retains all the features of Phusion Plus DNA Polymerase. In addition, two tracking dyes and a density reagent are pre-added to the master mix for direct loading of PCR products into gels.

High accuracy PCR enzymes are used for applications requiring high accuracy during DNA amplification such as cloning, sequencing or mutagenesis. Thermo Scientific Phusion high quality DNA polymerases were created by fusing a dsDNA-binding domain to a Pyrococcus polymerase-like proofreading enzyme. Thanks to this unique protein fusion engineering, Phusion DNA Polymerases have very low error rates, fast expansion rates, and high inhibitor tolerance.

Phusion™ Plus DNA Polymerase

The new Phusion Plus reaction buffer contains proprietary additives that stabilize primer/template duplexes during the annealing step at 60°C. This unique buffer formulation helps to save time and avoid errors in PCR studies by increasing PCR specificity without the need to optimize the annealing temperature for each primer pair.
Also, different PCR reactions can be cycled together using the same protocol with Phusion Plus DNA Polymerase and universal annealing.

Phusion Plus DNA Polymerase is inert at ambient temperatures due to the hot start mechanism mediated by Affibody molecules, allowing the coupled PCR reactions to be set up and stored at room temperature. Enzyme activity is fully restored after the first denaturation step.



To simplify PCR reaction setup, Phusion Plus DNA Polymerase is also available in ready-to-use master mix formats. Phusion Plus PCR Master Mix is a convenient 2X mix of enzyme, reaction buffer, magnesium chloride and dNTPs. Only template and primers need to be added to the PCR reaction. Phusion Plus Green PCR Master Mix also contains a density reagent and two tracking dyes for direct loading of PCR products into gels.
